Long-Term Cultivation of Human Cells (Chang) in Chemically Defined Medium and Effect of Added Peptone Fractions

چکیده
Freshly explanted tissue cells and most established cell lines cannot be cultivated in vitro indefinitely in chemically defined medium unsupple-mented with serum (1). Yeast extract and peptone the stimulatory components of either serum or peptone requires that the test system include no other unknown material. Thus, there is need for a chemically defined medium that will support con-TABLE I Fructose 1-6 phosphate 100.0 Fischer et al. 3.200 / Glucuronolactone 1.000 J 1-Homocystine 1.0 1 Ergothionine HCI 1.0 I Glucose 1-phosphate 1.0 [ Citric acid 1.0 ! Long-term cultivation of a human colony-stimulating factor-producing cell line in a protein-free chemically defined medium.
To examine whether a human colony-stimulating factor (CSF)-producing cell line, T3M-1, can propagate and secrete CSF without protein supplements, long-term cultivation of the cells was carried out in a protein-free chemically defined medium. By the use of stepwise decreases in the fetal bovine serum concentration, continuous growth of T3M-1 cells has been established in a protein-free F-10 medi...
متن کاملP-136: In Vitro Development of Ovine Oocytes Matured in A Chemically Defined Medium
Background: Despite great advances achieved in the field of in vitro oocyte maturation (IVM), almost all IVM media are supplemented with undefined component which not only pose the risk of pathogen transmission but also hinders our knowledge of molecular mechanisms of maturation. One of these components is serum that containing various concentration of unknown molecules affecting cellular proce...
متن کاملPeptone Supplementation of Culture Medium Has Variable Effects on the Productivity of CHO Cells
The optimization of cell culture conditions for growth and productivity of recombinant Chinese hamster ovary (CHO) cells is a critical step in biopharmaceutical manufacturing. In the present study, the effects of the timing and amount of peptone feeding of a recombinant CHO cell line grown in a basal medium in serum-free suspension culture were determined for eight peptones of different origin ...

Background: Culture media enrichment through the addition of protein hydrolysates is beneficial for achieving higher protein expression. Methods: In this study, designing the optimum mixture of four soy and casein-derived hydrolysates was successfully performed by design of experiment and specific productivity increased in all predicted combinations. Protein profile of recombinant CHO (rCHO) ce...
متن کاملLong-term cultivation and differentiation of human erythroleukemia cells in a protein-free chemically defined medium.
To examine whether a human erythroleukemia cell line, K-562, can proliferate and differentiate without protein supplements, long-term cultivation of the cells was carried out in a protein-free chemically defined medium. By the use of stepwise decreases in the fetal bovine serum concentration, continuous growth of K-562 cells was established in a protein-free F-10 medium. The cells have been pro...
